Then, I found out that I need to remove my ESC red wire as it is a BEC ESC. So, removing red wire form ESC is after ESC calibration ?

The 3s power can share with your esc.
Yes, remove the red wire and tape it up. That wire is live, you don't want to accidentally short it. Alternative get opto esc, it hac no bec.
If your radio can assign channel, what I did was to set all my channels to throttle and calibrate all the esc together but please remove all the prop before doing this.
